Metabolism of essential fatty acids. IV. Incorporation of linoleate into arachidonic acid.

چکیده
It has recently been demonstrated in this laboratory (1) that arachidonic acid is formed in the body from acetate and a Cl8 precursor and that, under the conditions of the experiment, linoleic acid is not synthesized from acetate. Spectrophotometric evidence (2, 3) has indicated that administration of linoleate to animals results in an increase in tetraene of the arachidonate type (as measured by alkali isomerization). The experiments reported in this paper were designed to test the hypothesis that the exogenous Cl8 precursor is linoleic acid and that the linoleate molecule is incorporated in its entirety into arachidonate.

O-16: Metabolism of Exogenous Fatty Acids, Fatty Acid-Mediated Cholesterol Efflux, PKA and PKC Pathways in Boar Sperm Acrosome Reaction
Background: For understanding the roles of fatty acids on the induction of acrosome reaction which occurs under association of cholesterol efflux and PKA or PKC pathways in boar spermatozoa, metabolic fate of alone and combined radiolabeled 14C-oleic acid and 3H-linoleic acid incorporated in the sperm was compared, and behavior of cholesterol and effects of PKA and PKC inhibitors upon fatty aci...
متن کاملThe role of pyridoxine in the metabolism of polyunsaturated fatty acids in rats.
Isotopic data reported by Mead, Steinberg, and Howton in 1953 (1) and by Steinberg, Slaton, Howton, and Mead in 1956 (2) firmly established the incorporation of acetate and of linoleate, respectively, into arachidonic acid in the rat. Witten and Holman in 1952 (3) had suggested that pyridoxine stimulated this conversion. They had shown that rats deficient in essential fatty acids and in pyridox...
متن کاملAn emerging risk factor for obesity: does disequilibrium of polyunsaturated fatty acid metabolism contribute to excessive adipose tissue development?
A positive energy balance (energy intake>energy expenditure), in which total fat intake plays an important role, is commonly regarded as a major factor contributing to obesity. Adipose tissue development, i.e. both size (hypertrophy) and number (hyperplasia), is stimulated by high dietary fat intake during early postnatal development, a susceptibility that now appears to continue well into adul...
متن کاملKinetics of Linoleic and Arachidonic Acid Incorporation and Eicosatrienoic Depletion in the Lipids of Fat-deficient Rats Fed Methyl Linoleate and Arachidonate.
Changes induced by dietary methyl linoleate and arachidonate in the fatty acid composition of liver phosphatidyl choline, phosphatidyl ethanolamine, cholesterol esters, and triglycerides were investigated in essential fatty aciddeficient rats. The esters were fed for 0, 24, 50, 96, 192, and 360 hr. The lipids were fractionated by thin-layer chromatography and the fatty acid composition was esti...

This study reports methods for the quantitative determination of stable isotope-labeled essential fatty acids (EFAs) as well as an experiment in which deuterium-labeled linoleic acid (18:2n-6) and alpha-linolenic acid (18:3n-3) were compared with those labeled with carbon-13 in rat plasma in vivo. Standard curves were constructed to compensate for concentration and plasma matrix effects. It was...
